Կալիֆոռնիայի Ռիվերսայդի համալսարանի գիտնականները պարզել են, որ PTPN2 գենի աշխատանքի խափանումը մեծացնում է աղիների աղիների բորբոքային հիվանդությանն (IBD, Inflammatory bowel disease) առնչվող վնասակար մանրէների նկատմամբ խոցելիությունը: Հետազոտության արդյունքները հրապարակվել են Gut Microbes բժշկագիտական հանդեսում:
Խոսքը AIEC մանրէների մասին է՝ Escherichia coli աղիքային ցուպիկների հատուկ տեսակ, որի մակարդակը բարձր է IBD-ից տառապող շատ բուժառուների շրջանում: Այդ մանրէներն ի վիճակի են կպչել աղիքային լորձաթաղանթին, ներթափանցել բջիջներ, վնասել պաշտպանիչ պատնեշը և մեծացնել բորբոքումը:
Նորմալ PTPN2 գենը կարևոր դեր է խաղում աղիների պաշտպանության գործում. այն օգնում է վերահսկել բորբոքային ռեակցիաները և պահպանել միկրոբիոմի հավասարակշռությունը: Սակայն IBD-ից տառապող մարդկանց մի մասի օրգանիզմում առկա է այդ գենի արատավոր տարբերակը, որը նվազեցնում է դրա ակտիվությունը: Արդյունքում, աղիքային պաշտպանական մեխանիզմները թուլանում են, և աղիքները ախտածին մանրէների նկատմամբ ավելի զգայուն են դառնում:
Հետազոտողները ցույց են տվել, որ PTPN2 –ի գործառույթների խաթարվում է, աղիքային բջիջները սկսում են արտադրել ավելի մասնագիտացված սպիտակուցներ իրենց մակերեսին: Այդ կառուցվածքները ծառայում են որպես «կպչման կետեր», որոնք նպաստում են հյուսվածքի մեջ AIEC-ի ամրակմանը և ներթափանցմանը: Այդ պաշտպանիչ պատնեշը կորցնելով՝ աղիքները այդ մանրէներին թույլ են տալիս բազմանալ և պահպանել խրոնիկական բորբոքումը։
Լրացուցիչ փորձերը ցույց են տվել, որ PTPN2-ը միաժամանակ մասնակցում է բնական հակամանրէային նյութերի արտադրությանը և պահպանում է լորձաթաղանթի ամբողջականությունը: Երբ գենը կանոնավոր է գործում, այդ մեխանիզմները սահմանափակում են ինչպես պայմանական ախտածին, այնպես էլ վնասակար մանրէների աճը։
Հեղինակները նշում են, որ IBD-ի համար արդեն օգտագործվող դեղամիջոցները՝ յանուս-կինազի ինհիբիտորները, կարող են մասամբ փոխհատուցել գենետիկ արատի ազդեցությունը: Լաբորատոր հետազոտություններում այդ դեղամիջոցները նվազեցրել են AIEC-ի աղիքային բջիջներ ներթափանցելու ունակությունը:
