Ենթաստամոքսային գեղձի քաղցկեղը վաղուց համարվում է քաղցկեղի բուժման ամենադժվար տեսակներից մեկը։ Այն գրեթե չի ենթարկվում թերապիայի ավանդական մեթոդներին և հաճախ դիմադրում է նույնիսկ առաջադեմ իմունաթերապևտիկ մոտեցումներին։
Հյուսիսարևմտյան համալսարանի հետազոտողները հայտնաբերել են մի նոր միջոց, որով ուռուցքը խուսափում է իմունային համակարգի հարձակումից. քաղցկեղի բջիջները ստեղծում են «շաքարային դիմակ», որը դրանք աննկատ է դարձնում օրգանիզմի պաշտպանիչ բջիջների համար։ Գիտնականները մշակել են նաև մոնոկլոնալ հակամարմին, որը կարող է քանդել այս քողարկումը։
Հետազոտության արդյունքները հրապարակվել են Cancer Research ամսագրում։
Լաբորատոր մկների վրա կատարված փորձերում այն «արթնացրել է» իմունային բջիջները՝ ստիպելով նրանց հարձակվել ուռուցքի վրա։
Գիտնականները պարզել են, որ քաղցկեղը նմանակում է «յուրային-օտար» ճանաչման բնական համակարգը։ Նորմալ բջիջների մակերեսը ծածկված է սիալաթթվով, որը Siglec-10 սպիտակուց-ընկալիչի միջոցով իմունային համակարգին ազդանշան է տալիս՝ «ինձ վրա մի հարձակվիր»։ Քաղցկեղի բջիջներն օգտագործում են նույն մեխանիզմը. ինտեգրին α3β1 սպիտակուցի վրա գտնվող սիալաթթուն թույլ է տալիս նրանց «յուրային» թվալ և խուսափել ոչնչացումից։
Արգելափակելով ինտեգրինի և ընկալիչի փոխազդեցությունը հակամարմինների օգնությամբ՝ հետազոտողները կարողացել են ակտիվացնել մակրոֆագերը և այլ իմունային բջիջներ, որոնք սկսել են արդյունավետորեն ոչնչացնել ուռուցքը։ Թերապիա ստացած կենդանիների մոտ ուռուցքի աճը զգալիորեն ավելի դանդաղ է ընթացել, քան վերահսկիչ խմբում։
Հետազոտության ավագ հեղինակ պրոֆեսոր Մոհամեդ Աբդել-Մոհսենը նշել է, որ այս մոտեցման մշակումը տևել է մոտ վեց տարի, և ընդգծել է, որ դրա ազդեցությունը դիտարկելը «մեծ առաջընթաց է»։
Թիմի հաջորդ քայլը մարդկանց մոտ հակամարմինների անվտանգության և դոզավորման փորձարկումն է, ինչպես նաև դրա օգտագործումը գոյություն ունեցող քիմիաթերապևտիկ և իմունաթերապևտիկ սխեմաների հետ համատեղ։
Զուգահեռաբար մշակվում է թեստ, որը թույլ կտա որոշել այն հիվանդներին, որոնց ուռուցքներն ունեն հենց այդպիսի «շաքարային քողարկում», որպեսզի թերապիան առավելագույնս արդյունավետ ուղղորդվի։
Այս աշխատանքը հեռանկար է բացում քաղցկեղի ամենաագրեսիվ տեսակներից մեկի դեմ նոր զենք ստեղծելու համար։ Ավելին, քողարկման մեխանիզմը կարող է ունիվերսալ լինել նաև այլ ուռուցքների, ինչպես նաև որոշ քրոնիկական բորբոքային հիվանդությունների համար, ինչը հետազոտության արդյունքները դարձնում է հատկապես խոստումնալից։
