Յեյլի համալսարանում անցկացված նոր հետազոտությունը ցույց է տվել, որ նեյրոնները՝ էներգատար բջիջները, որոնք կապում և ուղղորդում են ուղեղի ակտիվությունը, հագեցած են «պահեստային մարտկոցներով», որոնք գործի են դրվում նյութափոխանակային սթրեսի ժամանակ ուղեղի աշխատանքը պահպանելու համար։

Proceedings of the National Academy of Sciences ամսագրում հրապարակված հոդվածում գիտնականները նկարագրում են, թե ինչպես են նեյրոնները կուտակում իրենց սեփական գլիկոգենը՝ շաքարի ձև, որն օգնում է նեյրոններին կայուն մնալ, երբ նրանց էներգիայի հիմնական աղբյուրները շարքից դուրս են գալիս։

Ստացված արդյունքները ցույց են տալիս, թե ինչպես նեյրոնային բջիջները կարող են հարմարեցնել իրենց նյութափոխանակությունը և կարող են օգնել նյարդաբանական հիվանդությունների՝ ինսուլտի, նեյրոդեգեներացիայի և էպիլեպսիայի բուժման նոր մեթոդների մշակմանը. բոլոր այն խանգարումները, որոնց դեպքում դեր է խաղում էներգետիկ անբավարարությունը։

«Ավանդաբար համարվում էր, որ գլիալ բջիջները ծառայում են որպես «էներգետիկ պահեստներ»՝ պահելով գլիկոգեն և անհրաժեշտության դեպքում նեյրոններին վառելիք մատակարարելով», - բացատրել է հետազոտության համահեղինակ Միլինդ Սինգհը՝ Յեյլի բժշկական դպրոցի (YSM) բջջային կենսաբանության ամբիոնի դոկտորանտը։

Հետազոտության համար գիտնականներն օգտագործել են Caenorhabditis elegans (C. elegans) միկրոսկոպիկ կլոր որդը և գենետիկորեն կոդավորված HYlight ֆլուորեսցենտային բիոսենսորը, որը լուսարձակում է գլիկոլիզի (գործընթաց, որն օգտագործում են բջիջները՝ շաքարը տրոհելու համար՝ էներգիա ստանալու նպատակով) փոփոխություններին ի պատասխան։

Առաջընթացը տեղի ունեցավ, երբ հետազոտողները հայտնաբերեցին PYGL-1 ֆերմենտը՝ մարդկային գլիկոգենֆոսֆորիլազ ֆերմենտի որդանման տարբերակը, որը գլիկոգենը վերածում է նեյրոնների համար վառելիքի։ Երբ հետազոտողները հեռացրեցին PYGL-1-ը, որդի նեյրոններն այլևս չէին կարողանում էներգիա կուտակել թթվածնի ցածր պարունակությամբ սթրեսի պայմաններում։ Երբ ֆերմենտը հատուկ վերականգնվեց նեյրոններում, այս խափանումը վերացավ։

Այս երևույթը նկարագրելու համար գիտնականների թիմը հորինել է «գլիկոգեն կախյալ գլիկոլիտիկ պլաստիկություն» (GDGP) տերմինը։ Նրանք պարզել են, որ GDGP-ն հատկապես կարևոր է, երբ խանգարվում է միտոքոնդրիումների ֆունկցիան, օրինակ՝ հիպոքսիայի՝ թթվածնի սահմանափակ մատակարարման վիճակի ժամանակ։

Այս պայմաններում գլիկոգենը ծառայում է որպես արագ հասանելիության վառելիքի աղբյուր՝ օգնելով նեյրոններին ակտիվ մնալ, երբ մյուս համակարգերը կարող են կանգ առնել։

Նյութափոխանակային հարմարվողականության այս ունակությունը, որը հայտնի է որպես «գլիկոլիտիկ պլաստիկություն», օգնում է նեյրոններին պահպանել իրենց հիմնական գործառույթները սթրեսի պայմաններում։
